Today, my sister got fired and I got promoted to her job, got a nice payrise for it. My relationship with my sister will never be the same again. FML

Not your fault she got fired, and your boss has put their faith in you that you'll do the job better. If your sister is mad at you, tell her she shouldn't have done whatever it was that cost her the job.

if you don't really want a relationship with your sister then this is fine, congratulations, but if you do love your sister and want to maintain a relationship, you'll have to turn down the job.
